250 Bytes in Total - Close to 250 Characters
Most product categories only allow a total of 250 Bytes accross all backend keywords - ie. not 5 x 250 Bytes, but a total of 250 Bytes accross all 5 rows.
The keywords can therefore be spread across the 5 rows (eg. 50 per row), or summarized into the first row.
As soon as there are more than 250 Bytes, all of the keywords from the row which resulted in crossing the limitation will not not be indexed anymore.
Seller Central does not provide information on whether a product is only indexed for 250 bytes - this therefore has to be tested for each product (category), for example with the index checker mentioned below.
Special Letters - Multiple Characters
Spaces will not count as characters.
However, various special letters (eg. é) may cost multiple characters.

Sonar Keyword Index Checker
To make sure a specific product is indexed for certain keywords, you can use Sellics' free index checker at:
The index checker can also help determining whether a certaiin product category is affected by this change or not.
Seller Central Template for Backend Keywords
In the "Optimize keywords" function Sellics provides the option to summarize all monitored keywords into 250 Bytes or 5000 characters (1000 per row):
Cockpit > Monitored Products > Details > Keywords > Optimize keywords
Additionally, more space can be saved by using hyphens. This is explained further here.
